Trial Description

Purpose:

Octreotide and interferon alfa-2b may interfere with the growth of tumor cells and slow the growth of cancer. Monoclonal antibodies, such as bevacizumab, can block tumor growth in different ways. Some block the ability of tumor cells to grow and spread. Others find tumor cells and help kill them or carry tumor-killing substances to them. It is not yet known whether giving octreotide together with interferon alfa-2b is more effective than giving octreotide together with bevacizumab in treating patients with neuroendocrine tumor.

This randomized phase III trial is studying giving octreotide together with interferon alfa-2b to see how well it works compared with giving octreotide together with bevacizumab in treating patients with metastatic or locally advanced, high-risk neuroendocrine tumor.

Eligibility:

Eligibility criteria include the following:

• Measurable disease
• No metastatic cancer found only in bone, medullary thyroid cancer, islet cell carcinoma, or brain tumor
• At least 4 weeks since chemotherapy, radiation therapy, or surgery
• No more than one previous chemotherapy regimen
• No previous interferon, bevacizumab, SU11248, PTK/ZK, sorafenib, or pazopanib
• At least 3 weeks since octreotide

Final eligibility for a clinical trial is determined by the health professionals conducting the trial.

Treatment/Intervention:

Patients will be randomly assigned (have an equal chance of being placed) to one of two treatment groups.

Patients in group one will receive an intramuscular injection of octreotide and a 30- to 90-minute infusion bevacizumab on day 1.

Patients in group two will receive an intramuscular injection of octreotide on day 1. They will also receive an injection of interferon alfa-2b three times a week for 3 weeks.

Treatment in both groups may repeat every 3 weeks for as long as benefit is shown.

After finishing treatment, patients will be evaluated every 2-6 months for up to 3 years.
