Phase III Randomized Study of Letrozole With or Without Lapatinib Ditosylate in Postmenopausal Women With Stage IV Breast Cancer

Basic Trial Information

Trial Description

Purpose:

Estrogen can stimulate the growth of breast cancer cells. Hormone therapy using letrozole may fight breast cancer by blocking the uptake of estrogen. lapatinib may stop the growth of tumor cells by blocking the enzymes necessary for their growth. It is not yet known whether letrozole is more effective with or without lapatinib in treating stage IV breast cancer.

This randomized phase III trial is studying letrozole alone to see how well it works compared to letrozole and lapatinib in treating postmenopausal women with stage IV breast cancer.

Eligibility:

Eligibility criteria include the following:

• At least 18 years old
• No brain metastases
• No previous trastuzumab, cetuximab, gefitinib, or erlotinib
• No previous biological therapy, chemotherapy, hormone therapy, or radiation therapy for advanced or metastatic breast cancer

Final eligibility for a clinical trial is determined by the health professionals conducting the trial.

Treatment/Intervention:

Patients will be randomly assigned to one of two groups. Patients in group one will receive lapatinib and letrozole by mouth once a day. Patients in group two will receive a placebo and letrozole by mouth once a day. Quality of life is assessed before treatment begins, every 12 weeks during treatment, and after treatment ends. Treatment in both groups may continue for as long as benefit is shown.
