Phase I/II Study of Everolimus, Temozolomide, and 3D-Conformal Radiotherapy or Intensity-Modulated Radiotherapy Followed by Adjuvant Temozolomide With or Without Everolimus in Patients With Newly Diagnosed Glioblastoma

Basic Trial Information

Trial Description

Purpose:

Everolimus may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth and by blocking some of the blood flow to the tumor. Drugs used in chemotherapy, such as temozolomide, work in different ways to stop the growth of cancer cells, either by killing the cells or by stopping them from dividing. Radiation therapy uses high-energy x-rays to kill tumor cells. Giving everolimus together with temozolomide and radiation therapy may kill more tumor cells.

This phase I/II trial is studying the side effects and best dose of everolimus when given together with temozolomide and radiation therapy in treating patients with newly diagnosed glioblastoma.

Eligibility:

Eligibility criteria include the following:

• At least 18 years old
• At least 1 week, but no more than 6 weeks since previous surgery to remove the tumor or biopsy
• No oligodendrogliomas or oligoastrocytomas
• No previous chemotherapy for any brain tumor
• No previous temozolomide
• No previous radiation therapy to the cranium

Final eligibility for a clinical trial is determined by the health professionals conducting the trial.

Treatment/Intervention:

Patients will receive everolimus by mouth once a week in weeks 1-7 or weeks 1-8 and temozolomide by mouth once a day in weeks 2-7 or weeks 3-8. Patients will also undergo radiation therapy 5 days a week in either weeks 2-7 or weeks 3-8. Four to six weeks later, patients will receive everolimus by mouth once a week and temozolomide by mouth once a day in weeks 1-24. They will then receive everolimus by mouth once a week for as long as benefit is shown.

Patients undergo blood sample collection periodically for laboratory studies. Previously collected tumor tissue samples will be stored for future studies.

After finishing treatment, patients will be followed every 2 months for 1 year, every 3 months for 1 year, and every 6 months for 3 years.
