  • Olaparib in Treating Patients with Triple-Negative Non-metastatic Breast Cancer Who Have Completed Definitive Local Treatment and Chemotherapy

    This randomized phase III trial studies how well olaparib works in treating patients with triple-negative breast cancer that has not spread to another place in the body who have completed definitive local treatment and chemotherapy. Olaparib may stop the growth of tumor cells by blocking specific enzymes needed for the maintenance of deoxyribonucleic acid (DNA) repair. Olaparib may also enhance the DNA damaging effects of chemotherapy.

  • Pediatric MATCH: Targeted Therapy Directed by Genetic Testing in Treating Pediatric Patients with Relapsed or Refractory Advanced Solid Tumors, Non-Hodgkin Lymphomas, or Histiocytic Disorders

    This screening and multi-sub-study phase II trial studies how well treatment that is directed by genetic testing works in pediatric patients with solid tumors, non-Hodgkin lymphomas, or histiocytic disorders that have progressed following at least one line of standard systemic therapy and / or for which no standard treatment exists that has been shown to prolong survival. Genetic tests look at the unique genetic material (genes) of patients' tumor cells. Patients with genetic changes or abnormalities (mutations) may benefit more from treatment which targets their tumor's particular genetic mutation, and may help doctors plan better treatment for patients with solid tumors or non-Hodgkin lymphomas.

  • A Phase 2 Study of Cediranib in Combination with Olaparib in Advanced Solid Tumors

    This phase II trial studies cediranib maleate in combination with olaparib in treating patients with solid tumors that have spread to other parts of the body or cannot be removed by surgery, including breast cancer, non-small cell lung cancer, small cell lung cancer, and pancreatic cancer. Cediranib maleate and olaparib may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth. Cediranib maleate may also block the flow of oxygen to the tumor, and may help make the tumor more sensitive to olaparib.

  • Olaparib Treatment in Relapsed Germline Breast Cancer Susceptibility Gene (BRCA) Mutated Ovarian Cancer Patients Who Have Progressed at Least 6 Months After Last Platinum Treatment and Have Received at Least 2 Prior Platinum Treatments.

    Comparison of olaparib vs. physician's choice of single agent standard of care non-platinum based chemotherapy in patients with germline Breast Cancer susceptibility gene (gBRCA) mutated ovarian cancer who have progressed at least 6 months after the last platinum based chemotherapy. Patient should have received at least 2 prior lines of platinum based chemotherapy. The aim of the study is to assess the efficacy and safety of olaparib tablets.

Olaparib and onalespib may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth.

  • Selinexor with Multiple Standard Chemotherapy Regimens in Treating Patients with Advanced Malignancies

    This phase Ib trial studies the side effects and best dose of selinexor when given together with several different standard chemotherapy regimens in treating patients with malignancies that have spread to other places in the body and usually cannot be cured or controlled with treatment (advanced). Selinexor may stop the growth of cancer cells by blocking enzymes needed for cell growth. Drugs used in chemotherapy work in different ways to stop the growth of cancer cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Studying selinexor with different standard chemotherapy regimens may help doctors learn the side effects and best dose of selinexor that can be given with different types of treatments in one study.

  • Olaparib and Temozolomide with or without Irinotecan Hydrochloride in Treating Patients with Recurrent or Metastatic Ewing Sarcoma Previously Treated with Chemotherapy

    This phase I trial studies the side effects and best dose of olaparib and temozolomide in treating patients with Ewing sarcoma that has returned or spread to other places in the body after previous treatment with chemotherapy. Olaparib may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth. Drugs used in chemotherapy, such as temozolomide, work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Irinotecan hydrochloride may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth. Giving olaparib and temozolomide with irinotecan may be a better treatment for Ewing sarcoma.

  • Efficacy and Safety Study of Cediranib in Combination With Olaparib in Patients With Recurrent Platinum-Resistant Ovarian Cancer

    This is an open label, single arm, multi-center study to assess the efficacy and safety of the combination of cediranib and olaparib tablets in platinum-resistant relapsed high grade serous, high grade endometroid or clear cell ovarian, fallopian tube or primary peritoneal carcinoma patients who have received at least 3 prior lines of chemotherapy and who do not carry deleterious or suspected deleterious germline breast cancer susceptibility gene (BRCA) mutations.

  • Open-Label, Randomised, Multi-Drug, Biomarker-Directed, Phase 1b Study in Pts w / Muscle Invasive Bladder Cancer

    This is an open label, multi-drug, biomarker-directed, multi-centre, multi-arm, randomised Phase 1b study in patients with muscle invasive bladder cancer (urothelial) who have progressed on prior treatment. This study is modular in design, allowing evaluation of the safety, tolerability, pharmacokinetics and anti-tumour activity of multiple agents as monotherapy and as combinations of different novel anti-cancer agents in patients with muscle invasive bladder cancer. The study will consist of a number of study modules (substudies), each evaluating the safety and tolerability of a specific agent or combination.

Drugs used in chemotherapy, such as temozolomide, work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Giving olaparib and temozolomide may be a better treatment for recurrent small cell lung cancer.

  • Olaparib Tablets as a Treatment for Ovarian Cancer Subjects With Different HRD Tumor Status

    This is a non-randomized, open-label study to assess olaparib tablets as a treatment for subjects with different homologous recombination deficiency (HRD) tumor status and with platinum-sensitive, relapsed, high-grade ovarian cancer. Subjects should have received at least 2 prior lines of platinum-based chemotherapy.

  • Study of Pembrolizumab (MK-3475) Combination Therapies in Metastatic Castration-Resistant Prostate Cancer (MK-3475-365 / KEYNOTE-365)

    The purpose of this study is to assess the safety and efficacy of pembrolizumab (MK-3475) combination therapy in patients with metastatic castrate resistant prostate cancer (mCRPC). There will be three cohorts in this study with 70 participants enrolled in each cohort: Cohort A will receive pembrolizumab + olaparib, Cohort B will receive pembrolizumab + docetaxel + prednisone, and Cohort C will receive pembrolizumab + enzalutamide. Outcome measures will be assessed individually for each cohort.

    The purpose of the study is to learn from the real world practice of prescribing targeted therapies to patients with advanced cancer whose tumor harbors a genomic variant known to be a drug target or to predict sensitivity to a drug.   • Olaparib with and without AZD1775, AZD5363, and AZD2014 in Treating Patients with Advanced Solid Tumors

    This phase II trial studies how well olaparib works with and without other targeted therapies in treating patients with solid tumors that have spread to other places in the body. Olaparib, WEE1 inhibitor AZD1775 (AZD1775), Akt serine / threonine-specific protein kinase (Akt) inhibitor AZD5363 (AZD5363), and mammalian target of rapamycin (mTor) kinase inhibitor AZD2014 (AZD2014) may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth and repair. It is not yet known if giving olaparib alone or in combination with AZD1775, AZD5363, or AZD2014 will work better in treating patients with solid tumors that have spread to other places in the body.

  • Olaparib or Cediranib Maleate and Olaparib Compared with Standard Platinum-Based Chemotherapy in Treating Patients with Recurrent Platinum-Sensitive Ovarian, Fallopian Tube, or Primary Peritoneal Cancer

    This randomized phase III trial studies olaparib or cediranib maleate and olaparib to see how well they work compared with standard platinum-based chemotherapy in treating patients with platinum-sensitive ovarian, fallopian tube, or primary peritoneal cancer that has come back. Olaparib and cediranib maleate may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth. Cediranib maleate may stop the growth of ovarian, fallopian tube, or primary peritoneal cancer by blocking the growth of new blood vessels necessary for tumor growth. Drugs used in chemotherapy, such as carboplatin, paclitaxel, gemcitabine hydrochloride, and pegylated liposomal doxorubicin hydrochloride work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. It is not yet known whether olaparib or cediranib maleate and olaparib is more effective than standard platinum-based chemotherapy in treating patients with platinum-sensitive ovarian, fallopian tube, or primary peritoneal cancer.

  • Ascending Doses of AZD6738 in Combination With Chemotherapy and / or Novel Anti Cancer Agents

    This is a modular, phase I / phase 1 b, open-label, multicentre study of AZD6738 administered orally in combination with cytotoxic chemotherapy regimens and / or novel anti-cancer agents, to patients with advanced malignancies. The study design allows an investigation of optimal combination dose of AZD6738 with other anti-cancer treatments, with intensive safety monitoring to ensure the safety of the patients. The initial combination to be investigated is AZD6738 with carboplatin. The second combination to be investigated is AZD6738 with Olaparib. The third combination to be investigated is AZD6738 with MEDI4736.

  • Olaparib in Treating Patients with High-Risk, Biochemically-Recurrent Prostate Cancer Who Have Undergone Surgery

    This phase II trial studies how well olaparib works in treating patients with high-risk prostate cancer that has had a rise in prostate-specific antigen (PSA) after surgery. A rise in the PSA is a sign that there are tumor cells remaining in the body. Olaparib may help lower PSA and may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth.

    This partially randomized phase II trial studies how well abiraterone acetate, olaparib, and prednisone work in treating patients with prostate cancer that is resistant to hormones, has spread to other places in the body, and has deoxyribonucleic acid (DNA) repair defects. Abiraterone acetate and olaparib may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth. Drugs used in chemotherapy, such as prednisone, work in different ways to stop the growth of cancer cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Giving abiraterone acetate, olaparib, and prednisone may work better in treating patients with prostate cancer.

  • Olaparib and Ramucirumab in Treating Patients with Metastatic or Locally Recurrent Gastric or Gastroesophageal Junction Cancer That Cannot Be Removed by Surgery

    This pilot phase I / II trial studies the side effects and best dose of olaparib when given together with ramucirumab and how well they work in treating patients with gastric or gastroesophageal junction cancer that has spread to other places in the body, has come back, or cannot be removed by surgery. Olaparib may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth. Monoclonal antibodies, such as ramucirumab, may interfere with the ability of tumor cells to grow and spread. Giving olaparib and ramucirumab may work better in treating patients with gastric or gastroesophageal junction cancer.
